Medusa waits in the Holy Innocents' Cemetery, and beating her gives The Prophetess Arcana, the Petrifying Gaze spell and The Prophetess's Key. Never face her when she readies her stone gaze, drop off the platform when she swings in an arc, and hide behind fallen debris when she fires her multi-beam.
Kingy has not played the released game. The attack-by-attack strategy comes mainly from one written guide (GameTyrant), checked against the launch gameplay clips below; location and rewards come from two guides.

Where you fight her
Both guides put Medusa in the Holy Innocents' Cemetery. One walkthrough reaches her through the Roman Aqueduct, the Roman Baths and the Hot Springs, where you speak with a spirit and use a Mirror of Life before entering the Serpents' Lair, an arena of platforms above water.
A map guide names the fight area the Ossuary of the Innocents instead and lists the nearest Mirror of Life in the Ancient Sewers. Check your map if the names don't match.
Between attacks, stay on her flank rather than in front of her, land a few hits and back off before she turns. Keep enough room to dodge without falling into the water, and note that the Arcane Whip can't grapple onto her.
Stone gaze: when she pulls her fist straight back, turn to face away from her so you aren't looking when the ray fires (two guides). Another guide says to dodge sideways instead, and never jump or guard.Spoiler clip · Turning away from Medusa's stone gaze · reveal
On screen: Medusa, coiled on the left column, draws her fist back; the screen edges turn red and gaze lines sweep across the arena while Rose stands on the far right platform facing away from her.
Arc swing: when she pulls her fist back in an arc, drop off the platform to the ground, then use the whip hook to get back onto a platform before her spiked tentacle slams the ground.Spoiler clip · Medusa's arc swing, the ground tentacle and the hook back up · reveal
On screen: Medusa, coiled on her post, swings her fist in an arc over the platform while Rose waits on the ground below; a tentacle bursts from the ground, Rose hooks back up and hits Medusa from the platform.
Coil: when she wraps around a post she is switching sides. Move to the other side and attack as soon as she appears.Spoiler clip · Medusa switching sides to the other column · reveal
On screen: Medusa leaves the left column and dives into the water, rises at the far right and wraps around the right-hand column while Rose crosses the platforms toward her.
After you stun her, back away. She follows the stun with a long combo that hits several times.Spoiler clip · Medusa slumps onto the platform, then her coil attack while you keep your distance · reveal
When Medusa slumps onto the platform, get your hits in, then move away before she rears up. Wait on the far platform while her coils slam down and sweep through the middle of the arena.

Falling debris: when debris drops from the ceiling, get down behind it and wait there while she fires her multi-beam across the arena.Spoiler clip · Hiding behind fallen debris from Medusa's multi-beam · reveal
On screen: A chunk of debris falls beside the left platform, Rose drops down behind it, and Medusa's beams sweep across the arena above her before the debris crumbles.
Later in the fight she sends Medusa heads after you. Dodge them or hit them, and step around the small falling debris.Spoiler clip · Medusa heads in the air while you get back to her · reveal
When small Medusa heads start drifting across the arena, keep moving: whip-hook up toward a ring, then land on the platform beside Medusa and strike her face while she is up.

After the fight, pick up The Prophetess Arcana card. It unlocks Petrifying Gaze, a spell that turns enemies in front of you to stone.Spoiler clip · Picking up The Prophetess card after the fight · reveal
On screen: After the fight Rose stands on the arena platform under a glowing pickup, The Prophetess card fills the screen with Hold to equip and See Spell prompts, then Rose speaks about the card.
